Sorting out MIC, TRP, and CRAC Ion Channels

چکیده
A horde of cation-nonselective ion channel clans populate cell membranes. Their number is further augmented by removing extracellular calcium ([Ca 2 ] o ), turning well-behaved Ca 2 -selective channels into less discriminating sieves (Kostyuk and Krishtal, 1977; Almers and McCleskey, 1984). The Ca 2 ion stems the flood of ions moving through many cation channels by lodging in their throats, obstructing the path for monovalent ions by clinging to acidic groups in the narrow pore (Tsien et al., 1987; Hille, 2001). The trick of reducing [Ca 2 ] o uncovers channels whose conductances are inconveniently low in physiological solution. One of the most frustratingly low conductance Ca 2

Separation and Characterization of Currents through Store-operated CRAC Channels and Mg2+-inhibited Cation (MIC) Channels
Although store-operated calcium release-activated Ca(2+) (CRAC) channels are highly Ca(2+)-selective under physiological ionic conditions, removal of extracellular divalent cations makes them freely permeable to monovalent cations. Several past studies have concluded that under these conditions CRAC channels conduct Na(+) and Cs(+) with a unitary conductance of approximately 40 pS, and that int...
متن کاملDistinct Properties of CRAC and MIC Channels in RBL Cells
In rat basophilic leukemia (RBL) cells and Jurkat T cells, Ca(2+) release-activated Ca(2+) (CRAC) channels open in response to passive Ca(2+) store depletion. Inwardly rectifying CRAC channels admit monovalent cations when external divalent ions are removed. Removal of internal Mg(2+) exposes an outwardly rectifying current (Mg(2+)-inhibited cation [MIC]) that also admits monovalent cations whe...
متن کاملThe Drosophila Light-Activated Conductance Is Composed of the Two Channels TRP and TRPL
SUMMARY Drosophila phototransduction is a G protein-coupled, calcium-regulated signaling cascade that serves as a model system for the dissection of phospholipase C (PLC) signaling in vivo. The Drosophila light-activated conductance is constituted in part by the transient receptor potential (trp) ion channel, yet trp mutants still display a robust response demonstrating the presence of addition...
متن کاملCaT1 and the calcium release-activated calcium channel manifest distinct pore properties.
The calcium release-activated calcium channel (CRAC) is a highly Ca(2+)-selective ion channel that is activated on depletion of inositol triphosphate (IP(3))-sensitive intracellular Ca(2+) stores. It was recently reported that CaT1, a member of the TRP family of cation channels, exhibits the unique biophysical properties of CRAC, which led to the conclusion that CaT1 comprises all or part of th...
متن کاملOrai1 Mutations Alter Ion Permeation and Ca2+-dependent Fast Inactivation of CRAC Channels: Evidence for Coupling of Permeation and Gating
Ca(2+) entry through store-operated Ca(2+) release-activated Ca(2+) (CRAC) channels is an essential trigger for lymphocyte activation and proliferation. The recent identification of Orai1 as a key CRAC channel pore subunit paves the way for understanding the molecular basis of Ca(2+) selectivity, ion permeation, and regulation of CRAC channels. Previous Orai1 mutagenesis studies have indicated ...
